When we create an EC2 instance, tags are optional. In a company environment, can we make tags mandatory as to have more traceability ?

You can use Service control policies
https://docs.aws.amazon.com/organizations/latest/userguide/orgs_manage_policies_scps.html
and in your policies add Condition that requires a tag to perform an action
